Paracetamol’s Role in Ovarian Development Study

Researchers in Denmark have observed an unexpected trend in girls exposed to paracetamol (Tylenol) during pregnancy. This trend indicates potential differences in ovarian development markers from infancy into adolescence.

Study Findings on Paracetamol

Published in Human Reproduction Open, the study monitored 302 girls from a Danish pregnancy cohort. Researchers used detailed questionnaires and urine samples to track mothers’ paracetamol consumption during pregnancy. Data suggests daughters exposed to the drug may have smaller ovaries and uteruses. These differences were observed in ovarian activity and ovarian and uterine volumes.

Ovarian Reserve and Paracetamol

Girls exposed early in pregnancy showed reduced ovarian and uterine volumes compared to unexposed children. Later exposure correlated with fewer ovarian follicles. Additionally, lower anti-Müllerian hormone (AMH) levels, an indicator of ovarian reserve in adults, were noted.

Extended Observations

An independent cohort of 1,210 girls reinforced initial observations, showing smaller uterine volume at puberty and reduced ovarian volume during adolescence. Paracetamol use during pregnancy is common, with over half of pregnant women using the medication.

Concerns and Recommendations

Lead author Dr. Margit Bistrup Fischer clarifies that while findings do not prove causality, attention should be given to paracetamol use during pregnancy. Pregnant women should follow medical advice and use the lowest effective dose for the shortest time when necessary.

The Need for Further Research

Whether childhood differences affect fertility is still uncertain. Dr. Fischer suggests larger studies with longer follow-ups are needed to understand long-term impacts. Dr. Lucy Hooper emphasizes caution due to small subgroup sizes and precision issues in measuring certain differences.
